Protease inhibitors

Introduction

Protease inhibitors are molecules that inhibit the function of proteases, and many naturally occurring protease inhibitors are proteins. Protease inhibitors can be used as antiviral drugs and the FDA has approved the marketing of 7 protease inhibitor drugs including SQV, RTV, IDV, NFV, and APV, etc.

Protease inhibitors are further categorized into serine protease inhibitors, cysteine protease inhibitors, aspartate protease inhibitors, and metalloproteinase inhibitors based on homology of amino acid sequences and target proteases. Mutations in the key amino acids of proteases and their inhibitors can affect their physiological functions, stability, catalytic activity, inhibitory specificity, and so on.

It has become one of the current hotspots to explore various mutants of proteases and their inhibitors in nature, analyze their key active sites, and use protein engineering to modify and design proteases and their inhibitors with stronger activity, higher stability, better specificity, more environmentally friendly, and lower cost.
