GeneCopoeia Releases Multiplex Serological SARS-CoV-2 variants antigen microarray (64-plex) for SARS-CoV-2, the coronavirus responsible for the COVID-19 pandemic.

Purpose: GeneCopoeia has released the first multiplexed serological assay for SARS-CoV-2 (WT and variants), SARS-CoV-1, common coronaviruses, and other common respiratory-related viruses, to be used in antibody surveillance studies. This is the first highly multiplexed microarray-based assay for semi-quantitative detection of antibodies (IgG/IgM/IgA) against a whole spectrum of SARS-CoV-2 (wild-type and variant antigens),  other coronaviruses, and other common respiratory virus antigens in serum and plasma.

The OmicsArray™ COVID-19 SARS-CoV-2 and Variants Antigen Array (64-plex) is the first microarray-based multiplex assay to allow simultaneous detection of antibodies to the whole spectrum of  SARS-CoV-2 proteins (22 wild-type antigens, 19 variant antigens), other coronavirus species (SARS, MERS, 4 HCoVs) and other respiratory infection related viruses (influenza, RSV, CMV, etc.). Each slide contains 16 identical arrays enabling high throughput detection. The presence of Ig and anti-Ig as controls in each array allows the inter-array normalization and comparison among different samples. A high and low control sample is provided.

The 16-array slide format allows for simultaneous testing of up to 16 samples, and it’s easy to set up 4 plates (64 samples) on 1 rack for simultaneous detection of 56 antigen-specific antibodies for IgG, IgM, IgA, or other Ig subtypes in less than three hours, with as little as 2µl of sample, making it an efficient and extremely cost-effective solution for large-scale population studies. Most importantly, this array incorporates a large number of SARS-CoV-2 variant antigens, including the most contagious Alpha, Beta, Gamma and Delta variants, which are widely spreading around the world. We contend that as rapid changes occur in the virus and great challenges currently confront us, our high-throughput serological tests will provide powerful information to researchers and policymakers to help them better understand the spread of the novel coronavirus, and to evaluate the protective efficiency of COVID-19 vaccines in the community.

Technology overview

GeneCopoeia's OmicsArray™ antigen microarrays contain up to 120 purified proteins spotted onto nitrocellulose filters, which are adhered to glass slides. In addition, 8 spots are included for normalization. Each slide carries 16 identical arrays, and so can be used to process up to 15 samples simultaneously as well as a negative control. As little as 1 ul serum or 50 ul of other bio fluids are needed for each sample.

As shown in Figure 1, arrays are incubated with patient samples, and any antibodies in the  samples bind to their cognate antigens on the array. The arrays are washed to remove unbound antibodies and other proteins, then co-incubated with Cy3- and Cy5-labeled secondary antibodies. The dual labeling strategy is intended to distinguish between immunoglobulin (Ig) subtypes present within samples. For example. a Cy3-labeled anti-IgG secondary antibody is used to detect IgG antibodies, and a Cy5-labeled anti-IgM secondary antibody is used to detect IgM antibodies. Fluorophore-labeled secondary antobodies are available for detecting IgA, IgD, IgE, IgG and IgM immunoglubulins, as well as IgG subclasses IgG1, IgG2, IgG3, and IgG4.

After washing to remove unbound secondary antibodies, signals are detected using a GenePix® 4400A microarray scanner. The raw data is then analyzed using GenePix® 7.0 software.
