Phosphatases

Introduction

Phosphatases, which have the opposite function of protein kinases, remove phosphate groups from phosphorylated proteins by hydrolyzing the phosphate monoester into a phosphate group and a molecule with a free hydroxyl group. Dephosphorylation also occurs primarily on serine, threonine, and tyrosine residues. Members of the phosphoprotein phosphatases (PPPs) family include the PP1, PP2A, PP2B, PP4, PP5, PP6, and PP7 proteins, whose catalytic subunits are mostly combined with various regulatory subunits. In contrast, members of the metal-dependent protein phosphatases (PPMs) family, represented by PP2C proteins, do not have regulatory subunits and are dependent on metal ions such as Mn2+ or Mg2+ for their catalytic processes. Abnormalities in protein phosphorylation have been implicated in the development of a wide variety of diseases, ranging from cancer to inflammatory diseases, diabetes, infectious diseases, and cardiovascular diseases.
