GeneCopoeia provides the GLuc-ON™ HNF4 transcriptional response element (TRE) clone for analysis of the effects on genes regulated by hepatocyte nuclear factor 4 (HNF4). HNF4 is a transcription factor necessary for normal liver development, and its misregulation is associated with mature onset diabetes of the young type 1.
The GLuc-ON™ Oct4 TRE clone contains tandem repeats of an HNF4 binding site upstream of a minimal promoter and secreted Gaussia luciferase reporter gene, which allows robust and sensitive analysis without lysing the cells. The HNF4 response elements are engineered to provide strong activation with low background (Figure 1). The GLuc-ON™ HNF4 TRE clone is provided as transfection-ready DNA. Negative (non-activating), positive (constitutively-activated) and normalization (secreted alkaline phosphatase) control clones are also available for purchase.

HEK293a cells were co-transfected with HNF4TRE clone + GFP expression clone, negative control clone + HNF4 expression clone, or HNF4 TRE clone + HNF4 expression clone. Transfected cells were incubated for 48 hours. A Gaussia Luciferase assay was performed, and transcriptional response activity values are expressed as luminance fold activation.
