Introduction GeneCopoeia’s GLuc-ON™ MEF2 transcriptional response element (TRE) clone enables analysis of pathways that act through the myocyte enhancer factor 2 (MEF2) proteins. MEF2 is a family of 4 transcription factors (MEF2A, MEF2B, MEF2C, and MEF2D) in mammals that regulate genes involved in muscle development and cardiac stress. The GLuc-ON™ MEF2 TRE clone contains tandem repeats of an MEF2 binding site upstream of a minimal promoter and secreted Gaussia luciferase reporter gene, which allows robust and sensitive analysis without lysing the cells. The MEF2 response elements are engineered to provide strong activation with low background (Figure 1). The GLuc-ON™ MEF2 TRE clone is provided as transfection-ready DNA. Negative (non-activating), positive (constitutively-activted) and normalization (secreted alkaline phosphatase) control clones are also available for purchase.

Validation Data

    Figure 1. Activation of the GLuc-ON™ MEF2 TRE by treatment with phorbol 12-myristate 13-acetate (PMA). HEK293a cells were infected with Adenovirus particles expressing GFP and/or MEF2 clone, then treated with PMA for 24 hours. A Gaussia  Luciferase assay was performed, and transcriptional response activity values are expressed as luminance fold activation.

Introduction

GeneCopoeia provides the GLuc-ON™ TCF/LEF transcriptional response element (TRE) clone for analysis of the effects on Wnt signaling pathways mediated by TCF/LEF transcription factors, which recruit β-catenin to activate the promoters of several genes critical for function of the Wnt pathway. Wnt signaling plays fundamental roles in embryonic development, and its misregulation has been implicated in diseases such as cancer and type 2 diabetes.

    Figure 1. Activation of the GLuc-ON™ EGR1 TRE by treatment with phorbol 12-myristate 13-acetate (PMA). HEK293a cells were transfected with a negative control clone or the GLuc-ON™ EGR1 TRE clone. Transfected cells were incubated for 24 hours and then treated with varying amounts of PMA for an additional 24 hours. A Gaussia Luciferase assay was performed, and transcriptional response activity values are expressed as luminance fold activation.
