Introduction

GeneCopoeia’s GLuc-ON™ transcriptional response element (TRE) clones enable rapid and sensitive signal pathway analysis in mammalian cell lines. Using a secreted Gaussia Luciferase (GLuc) as the reporter, GeneCopoeia GLuc-ON™ TRE  clones are designed for robust and sensitive activation of specific signaling pathways in response to environmental stimuli or other experimental manipulation. Each GLuc-ON™ TRE clone comes either in transfection-ready or lentiviral format, and contains tandem repeats of a TRE placed upstream of a minimal CMV promoter and the Gluc gene.

Applications

GLuc-ON™ TRE clones offer powerful tools for several applications, including:

  • Drug discovery or validation. Identify small molecules that either stimulate or inhibit a signaling pathway of interest.
  • Analyze response of cells to proteins and peptides.
  • Analyze response of cells to hormones.
  • Functional genomics. Analyze the biological effects on a signaling pathway in response to gene activation, overexpression, knockdown, knockout, or mutagenesis (Figure 1).
  • Viral infection. Analyze response of cells to viruses.

 

Figure 1. Application example: Functional genomics using TRE clones. HEK293a cells were co-transfected with Oct4 TRE clone + GFP expression clone, negative control clone + Oct4 expression clone, or Oct4 TRE clone + Oct4 expression clone. Transfected cells were incubated for 48 hours. A Gaussia  Luciferase assay was performed, and transcriptional response activity values are expressed as luminance fold activation.

How do GLuc-ON™ TRE clones work?

GLuc-ON™ TRE response elements are engineered to provide strong activation of GLuc expression with low background. The GLuc reporter is a secreted protein, permitting convenient and rapid detection in the cell culture medium without lysing the cells (Figure 2).

Introduction GeneCopoeia provides the GLuc-ON™ ERSE transcriptional response element (TRE) clone for analysis of the response to endoplasmic reticulum (ER) stress resulting from the unfolded protein response. The ER stress response element (ERSE) is activated by several transcription factors, namely ATF6, to induce genes that express proteins that clear the cell of misfolded and unfolded proteins. In cases of prolonged ER stress, the pathway induces apoptosis. Malfunction of the ER stress pathway leads to diseases such as Huntingtin’s disease and diabetes.
The GLuc-ON™ MEF2 ERSE clone contains tandem repeats of an ERSE binding site upstream of a minimal promoter and secreted Gaussia luciferase reporter gene, which allows robust and sensitive analysis without lysing the cells.  The ERSE response elements are engineered to provide strong activation with low background (Figure 1). The GLuc-ON™ ERSE TRE clone is provided as transfection-ready DNA. Negative (non-activating), positive (constitutively-activted) and normalization (secreted alkaline phosphatase) control clones are also available for purchase.

Figure 1. Activation of the GLuc-ON™ ESRE TRE by treatment with thapsigargin. HEK293T cells were transfected with the GLuc-ON™ ESRE TRE clone or negative control clone. Transfected cells were incubated for 24 hours, then treated with different amounts of thapsigargin for 18 hours. Gaussia  Luciferase assay was performed, and transcriptional response activity values are expressed as luminance fold activation.

Introduction GeneCopoeia’s GLuc-ON™ JNK transcriptional response element (TRE) clone enables analysis of the effects on genes regulated by activator protein 1 (AP1). AP1 is a transcription factor complex comprised of the c-Fos and c-Jun family proteins,  and is necessary for signaling through the Stress-activated protein kinases (SAPK)/Jun amino-terminal kinases (JNK) (SAPK/JNK) pathway. The GLuc-ON™ JNK TRE clone contains tandem repeats of an AP1 binding site upstream of a minimal promoter and secreted Gaussia luciferase reporter gene, which allows robust and sensitive analysis without lysing the cells. The number of AP1 response elements has been optimized to maximize the sensitivity and minimize background levels (Figure 1). The GLuc-ON™ JNK TRE clone is provided as transfection-ready DNA. Negative (non-activating), positive (constitutively-activted) and normalization (secreated alkaline phosphatase) control clones are also available for purchase.
