Chemokines and chemokine receptors

Introduction

Cancer is a collection of diseases characterized by abnormal and uncontrolled cellular growth caused primarily by genetic mutations. Based on their role in cancer progression, driver genes can be classified into two main categories: oncogenes and tumor suppressor genes. Oncogene addiction refers to the phenomenon that certain tumours depend on one or more activated oncogenes, also known as driver oncogenes, to maintain their malignant biological phenotype.
Chemokines, or chemotactic cytokines, are a family of small cytokines or signaling proteins secreted by cells that induce directional movement of leukocytes, as well as other cell types, including endothelial and epithelial cells. In addition to playing a major role in the activation of host immune responses, chemokines are important for biological processes, including morphogenesis and wound healing, as well as in the pathogenesis of diseases like cancers.
Chemokines have been classified into four main subfamilies: CXC, CC, CX3C and C. Some chemokines are considered pro-inflammatory and can be induced during an immune response to recruit cells of the immune system to a site of infection, includes CCL14, CCL19, CCL20, CCL21, CCL25, CCL27, CXCL12 and CXCL13. This classification is not strict, for example, CCL20 can act also as pro-inflammatory chemokine. Others are considered homeostatic and are involved in controlling the migration of cells during normal processes of tissue maintenance or development. These chemokines are formed under pathological conditions (on pro-inflammatory stimuli, such as IL-1, TNF-alpha, LPS, or viruses) and actively participate in the inflammatory response attracting immune cells to the site of inflammation, such as CXCL-8, CCL2, CCL3, CCL4, CCL5, CCL11, CXCL10.
Chemokine receptors are G protein-coupled receptors containing 7 transmembrane domains that are found on the surface of leukocytes. Chemokine receptors associate with G-proteins to transmit cell signals following ligand binding. After receptor activation, the alpha- and beta-gamma-subunits of G protein dissociate to activate diverse downstream pathways resulting in cellular polarization and actin reorganization. Various members of small GTPases are involved in this process. Induction of nitric oxide and production of reactive oxygen species are as well regulated by chemokine signal via calcium mobilization and diacylglycerol production

Synaptic genes in autoimmune disease of nervous system

Introduction

The nervous system includes the central nervous system (CNS) and the peripheral nervous system (PNS). Viral infections, poisoning, genetic defects, immune damage, and metabolic disorders may trigger the development of neurological diseases. Neurological autoimmune diseases are diseases in which the body’s immune system mistakenly attacks its own nervous system, causing damage to the tissue structure and function of the nervous system. This kind of disease has a wide range of damage, and the clinical manifestations are complex and varied, which can involve the central nervous system, peripheral nervous system and nerve-muscle joints, resulting in neuronal or axonal damage, myelin loss, nerve-muscle joint destruction and other pathological changes.
Examples of CNS autoimmune diseases include optic neuromyelitis optica spectrum disease (NMOSD), myelin oligodendrocyte glycoprotein antibody-associated disease (MOGAD), multiple sclerosis (MS), acute disseminated encephalomyelitis (ADEM), autoimmune encephalitis (AE), and vasculitis of the CNS. These diseases have relatively independent clinical features, and some specific antibodies play an indispensable role in the diagnosis and differential diagnosis of the diseases. For example, patients with NMOSD may be positive for AQP4 antibodies, and patients with ADEM may be positive for MOG antibodies.
This clone set can be used for the development of biomarkers for neuroimmune diseases, expanding and improving the spectrum of humoral markers for neuroimmune diseases, which can be further used for differential diagnosis, disease course monitoring, efficacy and prognosis assessment, and drug target screening.

Cancer driver gene

Introduction

Based on their role in cancer progression, driver genes can be classified into two main categories: oncogenes and tumor suppressor genes. Oncogene addiction refers to the phenomenon that certain tumours depend on one or more activated oncogenes, also known as driver oncogenes, to maintain their malignant biological phenotype.
For example, EGFR gene mutation is a common mutation in lung cancer, which makes the EGFR receptor persistently activated and the signalling process aberrant, leading to long-term uncontrolled cell proliferation and growth, and becoming one of the driver genes of lung cancer cells. Cancer cells require driver oncogenes for sustained function, whereas normal cells do not.
Therefore, targeting oncogenes as therapeutic targets allows targeted drugs to specifically kill tumour cells without damaging normal cells. Martínez-Jiménez et al. present the Integrative OncoGenomics (IntOGen) pipeline, which applies to somatic mutations of more than 28,000 tumours of 66 cancer types reveals 568 cancer genes and points towards their mechanisms of tumorigenesis. These cancer driver genes’ identification has led to the development of the paradigm of targeted anticancer therapies and, more generally, to the search for genomic biomarkers of prognosis and response to treatments.

Product Information

UltraHiPF® DNA Polymerase is a heat-resistant ultra-fidelity DNA polymerase with 5′-3′ polymerase activity and 3′-5′ exonuclease activity. The polymerase can amplify uracil- and hypoxanthine- containing templates, GC- or AT- rich templates, genomes as low as one copy, and DNA fragments of more than 12 kb from different species. UltraHiPF® DNA Polymerase can specifically amplify 2 kb fragments at 1sec denaturation, annealing, and elongation, and can expand complex templates at 15-30 sec/kb. With Sanger sequencing, the mutation rate of this enzyme was 55 times lower than that of Taq DNA polymerase. The PCR product obtained by using the product has the Blunt-end.
