COVID-19 Coronavirus CRISPR and shRNA Solutions

Of the multiple host proteins required for SARS-CoV-2 pathogenesis, two of the most important are ACE2, the receptor that the viral spike protein uses for cell entry, and TMPRSS2, a serine protease required for S protein priming. COVID-19 Coronavirus Receptor Stable Cell Lines

General information
GeneCopoeia has produced HEK293T cell lines expressing ACE2 and TMPRSS2, two human host factors needed for efficient infection by SARS-CoV-2, the virus responsible for the COVID-19 pandemic.
These cell lines express ACE2 and/or TMPRSS2 mRNA at high levels on the cell surface, and are amenable to infection by viruses expressing SARS-CoV-2 Spike protein in their envelopes. Refer to the “Performance data” tab for more information.
ACE2-expressing cell lines are great for use with either native SARS-CoV-2 virus or SARS-CoV-2 Spike-pseudotyped lentiviruses for several applications, such as:
  • Vaccine development for the prevention of infection by SARS-CoV-2 virus
  • Studying the efficacy and mechanism of neutralizing antibodies against SARS-CoV-2 virus
  • Development of antiviral therapeutic agents

COVID-19 Coronavirus Lentiviral and AAV Solutions

General information

GeneCopoeia carries complete suites of state-of-the-art products for gene delivery using lentivirus and adenoassociated virus (AAV).

GeneCopoeia’s lentiviral solutions include:

  • Lentiviral plasmids. Available for expressing ORF cDNAs, gene promoters, miRNA precursors and inhibitors, CRISPR sgRNAs, and shRNAs for do-it-yourself, 3rd generation lentiviral packaging. Use the “Plasmids” tab for ordering information.
  • Ready-to-use particles. For infection of most mammalian cell types with either standard VSV-G pseudotyping, or for infection of ACE2-expressing cells with SARS-CoV-2 Spike glycoprotein pseudotyping. Lentivirus expressing SARS-CoV-2 viral proteins

Lentiviral particles for SARS-CoV-2 viral protein expression are available for 2 primary applications:
  1. Pseudotyping of lentivirus with the SARS-CoV-2 Spike (S) protein. Lentiviruses in common usage today are produced with the Vesicular Stomatis Virus G glycoprotein (VSV-G) expressed in the viral envelope, which enables the lentivirus to enter a wide range of mammalian cell types. However, during lentivirus packaging, the packaging plasmid encoding VSV-G can be replaced with a plasmid expressing SARS-CoV-2 Spike glycoprotein, which more narrowly limits their cell type infectivity to cells expressing ACE2, the human receptor for SARS-Cov-2 infection (Ou, X., et al., 2020. Nature Communications doi: 10.1038/s41467-020-15562-9; Hoffman, M., et al., 2020. Cell doi: 10.1016/j.cell.2020.02.052). These particles can be used for several applications, including vaccine development and testing antibodies that block cellular entry by SARS-CoV-2. The cyclophilins are a family of proline isomerases that function to aid in protein folding. Cyclophilins bind to the immunosuppressive drug cyclosporin A (CsA) and help mediate CsA’s pharmacological function. While there is no published evidence yet that cyclophilins are involved in the pathogenesis of SARS-CoV-2, several groups have shown that CsA can inhibit several viruses, including the virus that caused the 2002-2004 SARS pandemic, SARS-CoV, in a cyclophilin A (CypA)-dependent fashion (reviewed in Tanaka, Y., et al., 2013. Viruses 5, 1,250). Further, CypA interacts with SARS-CoV Non-structural protein 1 (Nsp1) as demonstrated in a yeast 2-hybrid screen (Pfefferle, E., et al.. 2011; PLOS Pathogens doi:  doi:10.1371/journal.ppat.1002331). SARS-CoV is highly similar to SARS-CoV-2, and, in fact, both viruses use ACE2 as their cellular receptor. Therefore, it is reasonable to include CypA in investigations of a potential role for CypA in SARS-CoV-2 pathogenesis.

FK506 binding proteins (FKBP) are, like cyclophilins, proline isomerases that bind to another immunosuppressive drug, FK506 and mediate its pharmacology. A recent publication in Nature (Gordon, et al., 2020) described the results of an affinity purification-mass spectrometry (AP-MS) screen that identified 332 human proteins that physically interact with 26 SARS-CoV-2 viral proteins. While it is unlikely that all 332 interactions are physiologically relevant, Gordon, et al. demonstrated that 66 of these human host proteins are potential targets of known drugs used for other purposes, and that a subset of these drugs inhibit viral replication in vitro.

Lentifect™ SARS-CoV-2 Spike-Pseudotyped Lentivirus

GeneCopoeia’s Lentifect™ SARS-CoV-2 Spike-pseudotyped lentivirus products produce a specially-packaged version of standard lentivirus, whereby the vesicular stomatitis G (VSV-G) envelope glycoprotein is replaced by the Spike (S) protein from SARS-CoV-2, the virus responsible for the COVID-19 pandemic. These particles, also known as “pseudoviruses”, are typically used to infect cultured cells expressing ACE2, the receptor for SARS-CoV-2 on multiple  tissue types (Figure 1).

Lentifect™ SARS-CoV-2 Spike-pseudotyped lentivirus and our ACE2- and TMPRSS2-expressing cell lines are great for several applications in a safe environment without the use of pathogenic SARS-CoV-2 viruses, such as:

SARS-CoV-2 Spike protein-pseudotyped lentiviral particles

GeneCopoeia’s Lentifect™ SARS-CoV-2 Spike protein-pseudotyped lentivirus is available as either premade, in vitro grade or custom-produced in vivo grade particles. Pseudoviral particles expressing Spike proteins from the original (“WT”) version as well as other variants, such as B.1.1.7 (first identified in the U.K.) and B.1.351 (first identified in South Africa) are available.

Spike variant mutational profiles

At the time of product introduction, GeneCopoeia’s R&D team used the GISAID database to determine the most frequently-occurring mutations in the Spike proteins from each variant to incorporate into our pseudovirus particles. These mutational profiles are indicated below. Note that each variant can have different sub-lineages, which could contain a slightly different repertoire of mutations from the variants that we have made available. Spike variant mutational profiles

At the time of product introduction, GeneCopoeia’s R&D team used the GISAID database to determine the most frequently-occurring mutations in the Spike proteins from each variant to incorporate into our pseudovirus packaging kits. These mutational profiles are indicated below. Note that each variant can have different sub-lineages, which could contain a slightly different repertoire of mutations from the variants that we have made available. If you would like SARS-CoV-2 Spike-pseudotyped lentivirus packaging kits expressing a different set of mutations than we have currently available. please contact inquiry@genecopoeia.com for a custom project.

HEK293T receptor cell lines expressing ACE2 and TMPRSS2

GeneCopoeia has produced HEK293T cell lines expressing human ACE2 alone, or ACE2 in combination with TMPRSS2, a protease that promotes Spike-ACE2 receptor interaction. These cell lines express ACE2 and/or TMPRRSS2 mRNA at high levels (Figure 1). Due to display of ACE2 protein (Figure 2) and/or TMPRSS2 protein ((Figure 3) on the cell surface, these cells are amenable to infection by viruses expressing SARS-CoV-2 Spike protein in their envelopes.
