CRISPR Plasmids
sgRNA expression plasmids are available for targeting virtually any gene in any experimental system. sgRNA plasmids express either sgRNA only or sgRNA + Cas9 nuclease in an all-in-one format. Lentiviral clones express sgRNA alone. Cas9-expressing lentiviral clones are available separately.
The CRISPR plasmid options listed below are intended for using 20 ntsgRNAs with the wild-type Cas9 nuclease from Streptococcus pyogenes(SpCas9). GeneCopoeia also provides alternate CRISPR options, such as Cas9 D10A nickase and other high-fidelity versions of Cas9, SaCas9, and shorter sgRNAs. Synthetic sgRNAs

GeneCopoeia offers customers a design and chemical synthesis service for sgRNAs (single-guide RNAs) targeting genes of interest. Chemically synthesised sgRNAs (including crRNA and tracrRNA) are purified via HPLC and modified by 2′-O-methyl and thiophosphate at the 3′ and 5′ ends. When Cas9 nuclease is present, the sgRNA recognises the target DNA sequence and guides Cas9 to cleave the target site. This creates double-strand breaks (DSBs) in the DNA and enables genome editing, including gene knockout, knock-in and mutation.

(A) Design of HUWE1-sgRNA and genomic PCR primers; (B) Transfection of HEK293T cells in a 6-well plate using HUWE1-sgRNA/Cas9 clones and sgRNA/Cas9 control clones (lane 2). Cells were harvested 40 hours after transfection, genomic DNA was extracted, and PCR amplification was performed using HUWE1-specific PCR primers. The PCR products were gel-purified, mixed with buffer, denatured, annealed, and then digested with T7 ENI for 60 minutes in a 37℃ water bath. The fragment length of the HUWE1 PCR product should be 520 bp, and the products after T7 ENI digestion should be 330 bp and 190 bp, respectively.

In a 24-well plate, Cas9-sgRNA complexes were transfected using CRISPR-Fectin™ transfection reagent (Cat# EF015), and cellular gDNA was sequenced 48 hours later.

Recombinant Cas9 Protein

Recombinant Streptococcus pyogenes Cas9 nuclease, purified from E. coli, is a great choice for genome editing applications in vitro or in vivo. Available in two versions, containing nuclear localization sequence (NLS), and either with or without a C-terminal 6× His-tag, at a concentration of 1 mg/ml. For optimal activity, use with GeneCopoeia’s CRISPR-Fectin™ Transfection Reagent.
All Cas9 nucleases are dissolved in storage buffer at 1 mg/ml.

The experimental group (lanes 1–4) consisted of HEK293T GFP stable cells co-transfected with a Cas9 expression clone and sgRNA clones targeting the p53, HUWE1, NCL3, and GFP genes, respectively. The control group (lanes 5–8) consisted of HEK293T GFP stably transfected cells co-transfected with plasmids expressing the Cas9 expression clone and a random sgRNA expression clone. Insertion-deletion mutations simultaneously present at each target site in genomic DNA were analyzed using T7 endonuclease assays. The bands marked with an asterisk (*) indicate that both the Cas9 nuclease and the sgRNAs targeting multiple sites effectively induced insertion-deletion mutations at the target sites (lanes 1–4). Sizes of PCR product bands and T7 endonuclease digestion products: GFP: 720 bp (full-length), 340 bp + 380 bp (digested); NCL3: 765 bp (full-length), 295 bp + 470 bp (digested); HUWE1: 520 bp (full-length), 190 bp + 330 bp (digested); P53: 825 bp (full-length), 475 bp + 350 bp (digested).

Product Information

GeneCopoeia’s OmicsArray™ Cancer and Neoplasm-associated Antigen Array is a protein microarray enabling powerful detection of autoantibodies that may be produced in cancer patients, especially those who have received immunotherapy. The array carries 120 superior-quality purified proteins spotted onto nitrocellulose filters, which are adhered to glass slides. Antigens known to be associated with cancers, as well as other autoimune conditions, are chosen based on a thorough review of peer-reviewed publications.

In addition to premade arrays, arrays containing customized sets of antigens are available, as well as array processing kits, array profiling services and data analysis. To order premade or custom arrays, please contact us.

Product Information

GeneCopoeia’s OmicsArray™ Brain and Neuronal-associated Antigen Array is a protein microarray enabling powerful detection of autoantibodies associated with neurological diseases. The array carries 120 superior-quality purified antigens spotted onto nitrocellulose filters, which are adhered to glass slides. Antigens known to be recognized by autoantibodies in patients with central nervous system disorders are chosen based on a thorough review of peer-reviewed publications. These autoantibodies could be either causes of neurological diseases or useful biomarkers.
