Immuno-oncology

Introduction

Immuno-oncology is a scientific field that studies the interactions between tumors and the immune system, revealing how tumors evade immune surveillance and elimination, as well as how they exploit the mechanisms of the immune system to promote their growth and spread. Cancer immunotherapy, developed on the theoretical foundation of immuno-oncology, represents a revolutionary approach to treatment. It activates or enhances the patient’s own immune system to recognize and destroy cancer cells, fundamentally differing from traditional cancer treatments such as surgery, radiation, and chemotherapy. Cancer immunotherapy comes in a variety of forms, including targeted antibodies, cancer vaccines, adoptive cell transfer, tumor-infecting viruses, checkpoint inhibitors, cytokines, and adjuvants.

Application

The Immuno-oncology clone set can be used in GSRA technology to perform high-throughput screening of various transmembrane or secreted proteins interacting with target proteins. The Genome-Scale Receptor Array (GSRA) is a semi-automated gene expression and detection system invented by Prof. Lieping Chen’s team, which is used to rapidly detect protein-protein interactions. This clone set fuels a number of research results. Screening of ligand proteins for LAG-3, revealing the role of this LAG-3-FGL1 pathway in tumour immunity and therapy[1]. Demonstrating that Siglec-15 inhibits antigen-specific T-cell responses in vitro and in vivo, and can serve as a potential target for cancer immunotherapy[2]. Identifying IGFBP7 interacts with its receptor CD93, and subsequently demonstrated that this interaction contributes to abnormal tumor vasculature[3]. And showing CD8α is critical for the maintenance of CD8+ T cells in a physiologically quiescent state in peripheral lymphoid organs[4].

Transcription factors

Introduction

Transcription factors, also known as trans-acting factors, are proteins that can bind to specific nucleotide sequences upstream of a gene, and these proteins can regulate the transcription of its genes, including TFIIA, TFIIB, TFIID, HSF, HIF, SREBP, and so on.
Based on protein structure, these transcription factors generally consist of DNA-binding domains, transcription regulation domains (including activation and repression domains), oligomerization sites, and nuclear location signals (NLS). Transcription factors are not only related to a variety of major human diseases. For example, p53, myc, ER, Id are related to cancer; PPAR are related to obesity; MEF2, HIF1 are related to heart disease.
Transcription factors are also involved in the regulation of plant growth and development, as well as stress defense responses. Studying the functions of transcription factors and their interacting factors is essential for understanding their roles in signaling cascade reactions, providing a theoretical basis for both basic research and production applications.

Secretory proteins

Introduction

Secretory proteins are proteins that are synthesized inside the cell and then secreted outside the cell to play a role, such as salivary amylase, pepsin, digestive enzymes, antibodies and some hormones. Protein secretion is one of the most important ways of information transfer between cells. General secretory proteins have a signal peptide at their N-terminus, which can be transported through the conventional secretory pathway and eventually secreted to the outside of the cell. These are conventional secretory proteins.
In recent years, it has been found that there exists another class of secretory proteins in the cell that can be secreted to the outside of the cell to play a role without signal peptides, and these proteins with unique secretion mechanisms are called unconventional secretory proteins. They are often used in the study of human body immunity changes caused by bacterial infections and their interaction mechanism with the host, which can provide a theoretical basis for the prevention of diseases and the development of targeted drugs.

Protease

Introduction

Proteases are widely distributed in animals, plants, and microorganisms, and their main role is to catalyze the hydrolysis of proteins and peptides. Proteases play an important role in organisms, and their activities are tightly regulated and controlled, because the overexpression of proteases could be detrimental to the health of the organisms. Together, proteases and their inhibitors are involved in the regulation of many physiological responses in the organism, such as the human pancreatic rennet protease (CTRC) that regulates the balance between trypsin activation and hydrolysis, the HIV-1 protease that plays an important role in viral replication, and the extracellular matrix metalloproteinase MT1-MMP mutation that affects bone tissue development.
Proteases are key factors in cellular health and viability and are involved in a variety of physiological processes such as replication, transcription, cell proliferation, differentiation, remodeling of the extracellular matrix, and processing of hormones and bioactive peptides. Proteases are implicated in many diseases, such as cancer, Alzheimer’s disease, arthritis, clotting disorders, allergies and infections.

Protease inhibitors

Introduction

Protease inhibitors are molecules that inhibit the function of proteases, and many naturally occurring protease inhibitors are proteins. Protease inhibitors can be used as antiviral drugs and the FDA has approved the marketing of 7 protease inhibitor drugs, including SQV, RTV, IDV, NFV, and APV, etc.
Protease inhibitors are further categorized into serine protease inhibitors, cysteine protease inhibitors, aspartate protease inhibitors, and metalloproteinase inhibitors based on homology of amino acid sequences and target proteases. Mutations in the key amino acids of proteases and their inhibitors can affect their physiological functions, stability, catalytic activity, inhibitory specificity, and so on.
It has become one of the current hotspots to explore various mutants of proteases and their inhibitors in nature, analyze their key active sites, and use protein engineering to modify and design proteases and their inhibitors with stronger activity, higher stability, better specificity, more environmentally friendly, and lower cost.
