Introduction

Phosphatases, which have the opposite function of protein kinases, remove phosphate groups from phosphorylated proteins by hydrolyzing the phosphate monoester into a phosphate group and a molecule with a free hydroxyl group. Dephosphorylation also occurs primarily on serine, threonine, and tyrosine residues. Members of the phosphoprotein phosphatases (PPPs) family include the PP1, PP2A, PP2B, PP4, PP5, PP6, and PP7 proteins, whose catalytic subunits are mostly combined with various regulatory subunits.
In contrast, members of the metal-dependent protein phosphatases (PPMs) family, represented by PP2C proteins, do not have regulatory subunits and are dependent on metal ions such as Mn2+ or Mg2+ for their catalytic processes. Abnormalities in protein phosphorylation have been implicated in the development of a wide variety of diseases, ranging from cancer to inflammatory diseases, diabetes, infectious diseases, and cardiovascular diseases.

Nuclear hormone receptors

Introduction

Nuclear hormone receptors refer to the target molecules of hormone action in the nucleus of the cell and belong to the nuclear receptors. Steroid hormone receptors and non-steroid hormone receptors are collectively referred to as nuclear hormone receptors.
Type I Steroid Receptors include: the estrogen receptor (ER), androgen receptor (AR), progesterone receptor (PGR), and glucocorticoid receptor (GR). Type II Non-steroid Receptors include: the thyroid hormone receptor (THR), the retinoic acid receptor (RARα), the vitamin D receptor (VDR), and the peroxidase proliferator activated receptor gamma (PPARγ). Receptors different from those on the cell surface are also known as nuclear hormone receptors.
Unlike receptors on the cell surface, these receptors are found only in postnatal animals (e.g., nematodes, insects, and vertebrates), and are mostly trans-acting factors that, when combined with the corresponding hormones, bind to cis-acting elements of DNA and regulate gene transcription, thus playing an important role in growth and development, metabolism, cellular differentiation, and many physiological processes in the body. Dysfunction of such receptors will lead to a series of diseases such as cancer, infertility, obesity, and diabetes.

Membrane bound proteins

Introduction

Membrane bound proteins are a class of proteins that can bind to cell membranes or other biological membranes to perform biological functions in the intra- and extracellular environments.
Membrane bound proteins have a relatively simple structure, usually consisting of one or more membrane-bound domains, an extracellular domain and an intracellular domain. Membrane-bound proteins are able to regulate cell morphology and movement, promote intercellular adhesion and interactions, and participate in biological processes such as cell signaling and material exchange by binding to biological membranes.
Numerous membrane proteins are involved in almost every life activity within the cell, including ligand-receptor binding, signal transduction, molecular transport, intercellular recognition and enzyme catalysis, etc. Therefore, membrane bound proteins are one of the important targets for many drug discovery and clinical therapeutic studies.

Introduction

Kinases are enzymes that catalyze the transfer of phosphate groups from high-energy phosphorus-containing molecules (e.g., ATP) to a substrate, and the largest kinase group is protein kinases. Kinases exert their effects by phosphorylating their respective downstream specific substrate proteins. They catalyze the transfer of γ-phosphate from ATP or GTP to the hydroxyl groups of serine, threonine, and tyrosine in substrate proteins or the glycosyl portion of lipids.
Kinases include: adenylate kinase (AMPK), creatine kinase (CK), pyruvate kinase (PK), hexokinase (HK), nucleoside diphosphate kinases (NDPKs), and thymidine kinases (TK1/TK2). Kinase dysregulation can lead to changes in enzyme activity and/or enzyme expression, subcellular localization, kinase stability, protein-protein interactions, etc. Aberrant regulation of kinase-mediated signaling pathways is responsible for all major human pathological conditions, such as cancer, cardiovascular diseases, neurodegenerative diseases, and metabolic disorders.
