Ion channels

Introduction

Ion channels are transmembrane proteins with special functions on cell membranes (including cytoplasmic and intracellular membranes). Since charged ions cannot freely pass through the lipid bilayer, they can only be transported through ion channels. Ion channels exist in the membranes of all cells, and their distribution and activity are very important for the excitability and function of cells and tissues.
A variety of ion channels exist in human tissues, and a variety of subtypes of each channel, such as calcium channels including members of KCTD10, KCNMA1, KCNIP2, SCN5A, and CHRNA3, potassium channels including members of TASK-1, TASK-3, TREK-1, MthK, and other members, each with specific nucleic sequences. They are both important factors in physiological regulation and can be used as targets for drug action to develop new specific drugs.

Histone modification enzymes

Introduction

Histone modification refers to the dynamic process by which histones are modified through methylation, acetylation, phosphorylation, adenylation, ubiquitination, ADP-ribosylation and other processes that are facilitated by specific enzymes.
Histone modifying enzymes include: histone acetyltransferase HAT and histone deacetylase HDAC for controlling acetylation; histone methyltransferases HMTs/KMTs, protein arginine methyltransferases PRMTs, and lysine demethylases KDMs for methylation; ubiquitylating histone ubiquitin ligases and deubiquitylating enzymes, etc. Histone modifications are a major form of epigenetics and play a crucial role in regulating genetic expression and phenotype. For example, histone modifications regulate the expression of nervous system-associated genes, which are crucial for the normal function of the nervous system, adaptive regulation, and the progression of neurodegenerative diseases, with distinct roles depending on the type of histone modification.

Histones

Introduction

Histones are alkaline proteins found in the chromatin of eukaryotic and prokaryotic cells, which form the structure of nucleosomes and are water-soluble together with DNA. They are mainly divided into five categories depending on their amino acid composition and molecular weight: H1/H5, H2A, H2B, H3, and H4.
Histones are rich in positively charged basic amino acids and can interact with the negatively charged phosphate groups in DNA. Histones have the functions of filling proteins and regulating chromatin, which is mainly realized through their modification.

GPCRs

Introduction

G protein-coupled receptors (GPCRs) are a large family of membrane protein receptors found only in eukaryotes. They are widely distributed in organs and tissues such as the central nervous system, immune system, cardiovascular system, and retina.
GPCRs that work under normal physiological conditions are mainly activated by ligand molecules and participate in the body’s development and normal functions. They are specific to multiple ligands, including GPR25, GPR35, TAS2R20, CRHR2, and OR6C74. However, if abnormal regulation of its related intracellular pathways occurs or exogenous pathogens attack body cells through them as receptors, it will lead to a series of diseases. Therefore, GPCRs are considered important drug development targets, with over 34% of modern drug targets being GPCRs.

Epigenetics

Introduction

Epigenetics is the study of heritable changes in gene function that do not involve alterations to the DNA sequence. Such changes often involve alterations that affect gene expression regulation, including DNA methylation, histone modifications, chromosome remodeling, non-coding RNA regulation, gene silencing, nucleolar dominance, transposable element activation, and dosage compensation effects of sex-related genes.

For example, epigenetic regulation of key genes related to neural development is an important regulatory mechanism that determines the differentiation of embryonic stem cells to neural cells.
