Drug target genes

Introduction

Drug targets are biomacromolecules in the body that have pharmacodynamic functions by interacting with drugs, such as certain proteins and nucleic acids. The genes that encode target proteins are also known as target genes. The identification of target molecules related to specific diseases in advance is the basis for modern drug development.
For example, CTLA4, a classical T-cell immune checkpoint in tumor immunotherapy, is a transmembrane protein encoded by the CTLA-4 gene, which is expressed in activated CD4+ and CD8+ T cells, as well as Lymphocyte Activation Gene 3 (LAG-3) and T-cell Immunoglobulin Mucin 3 (TIM-3).

SARS-CoV-2 host interaction genes

Introduction

SARS-CoV-2 is a strain of coronavirus that causes COVID-19. Studies have shown that more than 1,000 human proteins from different pathways are interaction partners of SARS-CoV-2. By interacting with host cell proteins, SARS-CoV-2 disrupts normal cellular functions and utilizes cellular equipment for replication. For example, the SARS-CoV-2 viral protein Nsp2 targets the translation initiation factor eIF4E2 to exploit the host translation machinery; ORF3a binds to VPS39 to inhibit autophagy; NSP12 promotes RIPK1 activation and thus stimulates ACE2 and EGFR receptors for viral replication.

CDs

Introduction

Cluster of differentiation (CD) antigens are cell surface molecules expressed on leukocytes and other cells associated with the immune system. Physiologically, CDs can act in a variety of ways, often acting as important receptors or ligands for cells that can initiate signaling cascades and thus alter cellular behavior, while some CD proteins are not associated with cellular communication but have other functions such as cell adhesion, cell activation, or cell inhibition.
For example, CD24 regulates B-cell proliferation and differentiation; CD274 is essential for T-cell proliferation and the production of γ-interferon and IL10, and its interaction with PDCD1 inhibits T-cell proliferation and cytokine production.

Cytokines and cytokine receptors

Introduction

Cytokines (CK) are low molecular weight soluble proteins (5-20 kDa) induced in a variety of cells by immunogens, mitogens, or other stimuli, generally through binding to the corresponding cytokine receptor on the cell surface, and through signaling, causing changes in cellular gene transcription.
Cytokines have a variety of functions such as regulating intrinsic and adaptive immunity, hematopoiesis, cell growth, APSC pluripotent cells, and repair of damaged tissues. Cytokines can be classified according to their main functions: interleukins (IL), colony-stimulating factor (CSF), interferon (IFN), tumor necrosis factor (TNF), transforming growth factor-β family (TGF-β family), growth factors (GF), and chemokine family.
Most cytokine receptors (CK-R) are heterodimers or multimers consisting of two or more subunits, usually including a specific ligand-binding alpha chain and a beta chain involved in signaling. In the natural state, cytokine receptors exist mainly in two forms: membrane-bound cytokine receptors (mCK-R) and soluble cytokine receptors (sCK-R) present in body fluids such as serum.
The complex biological activity of the cytokine is mediated primarily through binding to the corresponding mCK-R while sCK-R has a unique biological significance. Changes in sCK-R levels in relation to certain diseases have been increasingly emphasized by scholars, and some recombinant sCK-R genetically engineered products have been clinically validated.

Myasthenia Gravis (MG) Antigen Array, 68-Plex

Myasthenia Gravis (MG) is a chronic autoimmune neuromuscular disorder in which autoantibodies destroy the communication between nerves and muscle, resulting in weakness of the skeletal muscles. Myasthenia gravis affects the voluntary muscles of the body, especially those that control the eyes, mouth, throat, and limbs. MG is most often caused by antibodies to the acetylcholine receptors, but antibodies to other proteins, such as MuSK (Muscle-Specific Kinase), LRP4 (Lipoprotein-receptor-related protein 4), RYR (Ryanodine receptor), also can impair transmission at the neuromuscular junction.

GeneCopoeia’s OmicsArray™ Myasthenia gravis (MG) antigen array carries 68 MG-specific and/or -associate antigens. The 68-Plex MG Antigen Array is by far the largest collection of antigens for MG study and provides a powerful tool for highly sensitive profiling of autoantibodies in myasthenia gravis and associated diseases.

The 68 antigens included in the MG panel were chosen based on a thorough literature search. The superior-quality purified antigens were coated onto the 3D surface of microchips to ensure that antigens retain their natural conformations. Internal Ig and anti-Ig controls are included in each array for normalization and quantification purposes. Additionally, the utilization of fluorescent 2nd antibodies in our microarrays allow for the detection of different types and sub-types of antibodies (IgG1, IgG2, IgG3, IgG4, IgM, IgA) per experiment, minimizing the requirement of sample needed.
