GeneCopoeia’s Lentifect™ purified and ultra-purified lentivirus for in vitro and in vivo use are available either custom-made or pre-made. They are engineered for the transduction of nearly all mammalian cells, including difficult-to-transfect cells. To ensure the highest possible titer and quality, Lentifect™ lentivirus is produced from standardized protocols, with stringent quality control processes, which include sequence verification of the insert and qRT-PCR based titer estimation.

Lentiviral vectors are potent vehicles for delivery of genes into a wide range of cell types including difficult-to-transfect cells. Lentiviral particles transduce cells and integrate into the host genome in dividing and post-mitotic cells resulting in long-term expression of the transgene in vitro. Producing, concentrating and titrating lentiviral particles, however, is time consuming and requires some experience to achieve high titers and consistent results. GeneCopoeia’s Lentifect™ purified lentiviral particles save you time and effort, and provide high-level expression of genetic elements of interest (Figure 1).

Introduction

The Lenti-Pac™ Lentivirus Concentration Solution is designed for quick and simple concentration of lentiviral particles. The concentration is accomplished by mixing the proprietary reagent with the lentiviral supernatant, followed by a short incubation then centrifugation with an ordinary centrifuge.

Figure 1. Concentration of lentiviral particles. Lentiviral particles expressing GFP (2×10⁷ TU/ml) were concentrated 10 times using Lenti-Pac™ lentivirus concentration solution. The concentrated lentiviral particles were used to transduce H1299 cells in a 24-well plate. The pictures were taken 48 hours after the transduction. A: 0.2 µl of Standard lentivirus; B: 0.02 µl of concentrated lentivirus (incubated 4 hours with the concentration solution before centrifugation). Introduction

GeneCopoeia’s Lenti-Pac™ HIV Expression Packaging Reagents are engineered for efficient, do-it-yourself lentiviral particle production. When combined with GeneCopoeia lentiviral constructs, the results are high viral titers and robust expression levels. We offer four Lenti-Pac™ Lentiviral Packaging Reagents, each available separately.

Lenti-Pac™ HIV expression packaging kit. Comes with an optimized lentiviral packaging plasmid mix, eGFP positive control plasmid, EndoFectin™ Lenti, a transfection reagent optimized for virus production, and TiterBoost™ reagent, which increases titers 5-10 fold. This 3rd-generation packaging system ensures high-titer production of particles that are incapable of self-replication.
Lenti-Pac™ 293Ta packaging cell line. Enables easy and efficient lentiviral plasmid transfection for high-level expression of viral proteins required for packaging.
Lenti-Pac™ concentration solution. Designed for quick and simple concentration of lentiviral particles. Concentration is achieved by mixing the proprietary reagent with the lentiviral supernatant, followed by a short incubation and centrifugation.
Lenti-Pac™ titration kit. Designed for quick and simple titration of lentiviral particles. Used for etermining viral copy number and to confirm the success of packaging reactions. Includes reagents for RNA extraction and qRT-PCR, as well as a standard control.
Lenti-Pac™ Lentivirus Storage Buffer. Using this product to dissolve lentivirus particles can improve the stability of lentivirus at -80°C and effectively preserve functional viral particles during repetitive freeze-thaw cycles, effectively slowing down the decline in virus titer and preserving lentiviral infectivity. It is suitable for resuspending lentivirus after ultracentrifugation, polyethylene glycol precipitation (PEG precipitation), and ultrafiltration concentration.
