Database for Human Recombinant Proteins Produced with OmicsLink™ ORF Cell Free Expression Clones and RTS Wheat Germ CECF System

Introduction

A collaborative project was jointly conducted and completed by Roche Applied Science (RAS), GeneCopoeia and FulenGen. The aim of this project was to combine the material resources and technology know-how's owned by FulenGen, GeneCopoeia, and RAS, with the purpose of developing and providing researchers with a plug and play solution for recombinant protein expression, production and modification that is simple, quick, reliable, economical. The following specific goals of this project were set and met at high success rate.

  • Construct and optimize one or more expression vectors for use in large scale screening test using pIVEX vector backbone and OmicsLink™ ORF clones
  • To construct and adapt a collection of 13,000 OmicsLink™ human full length ORF expression clones so that they are fully compatible with RAS's RTS Wheat Germ Cell Free protein expression system, including the reagent kits and instrument.
  • Reactions of protein expressions are performed using the RTS Wheat Germ CECF kits and instrument with these specially constructed expression clones.
  • Proteins produced using clones and RTS WG system are assayed and analyzed, including SDS PAGE/Western Blot analysis.

Database

This database contains protein expression information of 13,000 human recombinant proteins produced using RTS wheat germ CECF system and OmicsLink™ ORF expression clones. The information includes 1) protein yield, solubility, and molecular size information of each tested protein assayed by SDS gel electrophoresis with Comassie-blue staining and Western Blotting using anti-tag antibody; 2) detailed annotation of each test protein, including protein family or group affiliation; 3) experimental conditions and reagent usage information. A web user interface was built for easy search of proteins of interest by users.

Expression Data Availability

The result of this expression screening is being published in a peer-reviewed journal, and not yet available to external users. Interested researchers are encouraged to contact support@genecopoeia.com for access rights and conditions. Proteins and their expression information in the database are organized and presented in many ways. The table below shows an example of the data organized in major protein families and groups that are considered as rich in drug targets and therapeutic agents.
