Product Information AccelerRT^® Ⅲ Reverse Transcriptase (RT) is an engineered version of M-MuLV RT with reduced RNase H activity. RNase H specifically degrade the RNA stand in RNA-DNA hybrids, therefore it probably degrade the RNA template in RNA-DNA hybrids when the first-stand cDNA synthesis. The enzyme can be used to synthesize more full-length product than conventional M-MLV, stronger extension ability. And it can used to Real Time RT-PCR.

Product Information

2× UltraHiPF® PCR Mix is a 2× premixed solution containing UltraHiPF® DNA Polymerase, dNTPs, and all other components required for PCR, excluding DNA templates and primers. The premixed solution saves time and reduces contamination by reducing the number of pipetting steps required to prepare the PCR reaction system.

UltraHiPF® DNA Polymerase is a heat-stable, ultra-fidelity DNA polymerase with 5′-3′ polymerase activity and 3′-5′ exonuclease activity. The polymerase can amplify uracil- and hypoxanthine- containing templates, GC- or AT- rich templates, low copy genomes, and DNA fragments greater than 12 kb in length from various species. The enzyme has a 55-fold lower mutation rate than Taq DNA polymerase by Sanger sequencing. The PCR products obtained with the product have blunt ends.

Product Information

UltraHiPF® DNA Polymerase is a heat-resistant ultra-fidelity DNA polymerase with 5′-3′ polymerase activity and 3′-5′ exonuclease activity. The polymerase can amplify uracil- and hypoxanthine- containing templates, GC- or AT- rich templates, genomes as low as one copy, and DNA fragments of more than 12 kb from different species. UltraHiPF® DNA Polymerase can specifically amplify 2 kb fragments at 1sec denaturation, annealing, and elongation, and can expand complex templates at 15-30 sec/kb. With Sanger sequencing, the mutation rate of this enzyme was 55 times lower than that of Taq DNA polymerase. The PCR product obtained by using the product has the Blunt-end.

Product Information AccelerRT® 5G Template Switching RT Enzyme Mix (RNase H-) is a novel RT enzyme that was evolved in vitro from MMLV RT. The enzyme possesses RNA- and DNA-dependent polymerase activities but lacks RNase H activity. Its efficient template-switching function allows it to be used for full length cDNA products. The engineered enzyme has greatly improved thermal stability, processability and synthesis rates compared to the wild type MMLV RT enzyme.

Template Switching function

Figure 1. Introduction to the principle of the Template Switching RT Enzyme. The Oligo(dT) VN Primer is used to synthesize the first-strand cDNA. Upon reaching the 5′ end of the RNA template, a few non-template nucleotides are added to the 3′ end of cDNA (Template-switching) using the terminal deoxynucleotidyl transferase (TdT) activity of reverse transcriptase. The second-strand of the cDNA is synthesized using a template-switching oligo. Finally, the full-length cDNA product is obtained by PCR amplification using reverse gene-specific primers and forward Template-switching oligo (TSO)-specific primers.

Technology overview

After washing to remove unbound secondary antibodies, signals are detected using a microarray scanner (e.g., GenePix® 4000B, InnoScan 710, or equivalents). The raw data is then be analyzed using GenePix® Pro 7.0, or Mapix software.

Data analysis

1. Collection of raw data. After scanning, raw signal intensities are collected in an Excel file, as shown below.
