Lenti-Pac™ Lentiviral Packaging Kits — efficient transduction, high
titers and high levels of protein expression
The GeneCopoeia Lenti-Pac™ HIV and FIV Expression Packaging Systems includes an optimized lentiviral packaging plasmid mix, eGFP control clone and a new transfection reagent developed to work with lentiviral-based constructs. When combined with GeneCopoeia OmicsLink™ ORF Lentiviral Constructs the results are high titers and robust expression levels.
Both the HIV and FIV Expression Packaging Systems safely ensure efficient expression of recombinant transcripts in mammalian cells. A comprehensive, optimized system provides high-quality expression constructs, packaging plasmids for lentiviral-based transduction and a new reagent for efficient transfection into packaging cells.
Fully-sequenced, ready-to-use and validated expression constructs (OmicsLink lentiviral-based protein encoding ORFs, miRNAs and shRNAs) improve vector function for very effective transduction and ensure high-level expression in dividing and non-dividing mammalian cells.
Packaging plasmids are designed for coordinated, high-level expression of all essential lentiviral packaging elements to ensure efficient packaging of the recombinant transcript into infectious virus particles, as well as for preventing the viral sequences from self-replicating.
EndoFectin™ Lenti, a proprietary transfection reagent, has been specifically optimized and tested with several lentiviral control clones, including eGFP and mCherry, for co-transfection of lentiviral expression constructs and packaging plasmids into packaging cells to deliver high lentiviral particle titers (Figure 1).
When used with the GeneCopoeia optimized packaging system, GeneCopoeia lentiviral clones produce infectious supernatants that are typically ready without concentration to use for highly efficient target-cell transduction (Figure 2).

Figure 1.HEK293T packaging cells were transfected with GeneCopoeia Lentivirus Vector, Ex-mCHER-Lv105 (which expresses mCherry fluorescent protein) and Lenti-Pac HIV packaging Mix using either EndoFectin Lenti (left panel), or a leading lipid-based transfection reagent (right panel). Expression of mCherry and cell health were checked 48 hours post-transfection using fluorescence microscopy (upper panel) or phase contrast microscopy (lower panel). The titer of lentivirus generated with EndoFectin Lenti reagent was over 20-fold higher than with the lipid-based transfection reagent.
