The ORFEXPRESS™ Gateway® Shuttle Clone product line currently contains over 40,000 unique clones derived from full length human and mouse genes that were chosen through a strict selection process. This process included the extraction, comparison, and validation of gene sequences and annotation information from multiple public and private sources; clustering and reduction of redundant gene sequences; filtering out error sequences or sequencing errors, plus several other manual curation steps. Once this stringent selection process was completed, the entire coding open reading frames (ORFs) of these genes were generated from sequence validated full length cDNA clones or high quality human tissue cDNA libraries and then inserted into modified Gateway® Entry Vector using GeneCopeia's proprietary high fidelity cloning technologies. These ORFs were manufactured to be compatible with Invitrogen Corporation's Gateway® Technology, an efficient universal cloning system developed by Invitrogen Corp.
The modification in the linker between attachment site (attL1 site) and start codon (ATG) of full ORF makes ORFEXPRESS™ Gateway® Shuttle Clones compatible with other cloning systems using the classical restriction-ligation cloning method.
All clones were validated through a rigorous 2-step quality control and assurance process:
1) After cloning, each ORF clone is fully sequenced;
2) Before delivering to customer, the clone is validated again by PCR with the gene specific primers.
Compatibility of ORFEXPRESS™ Gateway® Shuttle Clones with Invitrogen Corporation's Gateway® Cloning Technology makes it possible the rapid and simple transfer of the coding ORFs into any Gateway® destination expression vector for expression and functional analysis of target proteins in many hosts such as E.coli, yeast, baculovirus, CHO and mammalian cell lines as well as cell free transcription and translation coupling systems.
