ORFEXPRESS Gateway® PLUS Shuttle Clones — a high value choice for both traditional and recombination cloning

ORFEXPRESS Gateway® PLUS Shuttle clones offer both recombination and multiple cloning sites making them highly
multifunctional and cost-effective. The collection consists of 20,000 human and 15,000 mouse Gateway PLUS shuttle clones.
Compatibility of ORFEXPRESS™ Gateway PLUS shuttle clones with Invitrogen Gateway cloning technology makes the
transfer of protein-coding ORFs into any Gateway destination expression vector simple and rapid. These clones can then be used
for expression and functional analysis of target proteins in E.coli, yeast, baculovirus, CHO and mammalian cell lines as well
as cell-free transcription and translation coupling systems.
Features of Gateway PLUS Shuttle Clone
- Available with and without stop codons
- Ribosomal binding sites (Shine Dalgarno and Kozak
Sequence) for optimal translational context
- Multiple cloning sites included

Recombination cloning – The presence of attL1 and attL2 sites enables transfer via recombination of ORFs into any destination
vector using Gateway technology.
More compatibility − Unlike other Gateway clones, Gateway PLUS enable traditional restriction digestion ligation cloning. The full ORF of Gateway
PLUS shuttle clones have multiple cloning sites in the linker between attachment site (attL1 site) and start codon (ATG). This
makes them compatible with many other cloning systems using the classical restriction-ligation cloning method.
Rigorous quality control and assurance process
Human and mouse Gateway PLUS shuttle clones are derived from full-length human and mouse genes that were chosen through a
strict selection process. This process includes the extraction, comparison as well as validation of gene sequences and
annotation information from multiple public and private sources; clustering and reduction of redundant gene sequences,
filtering out error sequences or sequencing errors plus several other manual curation steps.
Following this stringent selection process, the entire coding open reading frames (ORFs) were generated from sequence
validated full-length cDNA clones or high-quality human tissue cDNA libraries and then inserted into modified Gateway® entry
vector using GeneCopoeia's proprietary high – fidelity cloning technologies.
Once the clone is constructed, GeneCopoeia follows additional quality control steps to ensure the right clone is delivered
to you.
- All ORFs are fully sequenced.
- PCR amplification and size validation
- Enzyme digestion check of the integrity of whole plasmid.
